Development of mass spectrometry imaging methods using LESA-HRMS

Have you ever wondered what happens to a drug you take as a tablet, how it is distributed in the body and how it finds its destination to be effective?

The global biotransformation team supports all Novartis scientists worldwide in all disease areas and across all technology platforms (e.g. low molecular drugs, peptides, siRNA or cell and gene therapies) with information on drug metabolism and distribution, contributing to the development of effective and safe new medicines.

We wish to expand our capabilities in the field of mass spectrometric imaging (MSI) and are looking for a highly motivated and curious Master student who would like to develop first methods for Liquid Extraction Surface Analysis (LESA)-MSI applications as part of their Master's thesis. MSI is a powerful and modern technology that helps to understand drug metabolism and distribution in tissues to guide pharmacokinetic and pharmacodynamic assessments. You will be responsible for setting up a new LESA system in combination with an HRMS instrument that is reserved for this project.

This position will offer you the unique opportunity to apply and deepen your knowledge in the field of (bio)analytical chemistry using state-of-the-art equipment (especially HPLC and MS). You will take your first steps in independent research, working on innovative science under the guidance of experienced scientists in a highly dynamic, international environment.

Your responsibilities:
• Set-up a brand new analysis (LESA) imaging system in combination with a micro-HPLC system and a high resolution mass spectrometer (Thermo Fisher Orbitrap Elite)
• Optimize and validate the LESA-system with a test set of small molecule reference compounds and animal tissues or tissue surrogates
• Evaluate possible strategies for the quantitative assessment of drug metabolites
• Extend the use of this technique to study distribution of biotherapeutics (like antibody drug conjugates or oligonucleotides) in tissues
• Apply developed LESA system to real life in vivo samples obtained from ongoing projects and support the discovery and development of new, effective and safe drugs

Minimum Requirements

• Student currently enrolled in a Masters degree in a Life Science discipline (e.g. Pharmacy, Chemistry, Food Chemistry, Biotechnology, Biochemistry or Bioengineering etc.)
• Good theoretical and scientific knowledge in the area of liquid chromatography (e.g. UHPLC, RP-HPLC), mass spectrometry (e.g. HRMS, MS/MS) and in sample preparation of biological fluids (e.g. SPE, LLE, protein digestion, derivatization) and the openness to learn new techniques and deepen your knowledge
• Initial experience and knowledge in the field of mass spectrometry imaging (e.g. MALDI or DESI) and the use of specific visualization software are an asset
• You have a creative mindset, a self-driven attitude, a high level of learning agility, you are eager to expand your knowledge and above all you value collaboration in teams
• Proficiency in English, with German skills are advantageous
